Tóm tắtGiới thiệuHen suyễn là một bệnh hô hấp mãn tính phổ biến ở trẻ em và người lớn. Một yếu tố di truyền quan trọng ảnh hưởng đến nguy cơ mắc hen suyễn đã được nhận diện từ lâu, mới đây thông qua việc xác định nhiều gen (ví dụ: ORMDL3, PDE4D, HLA‐DQ, và TLE4) thông qua các nghiên cứu liên kết toàn bộ gen.Mục tiêuXác định các biến thể di truyền liên quan đến trạng thái mắc hen suyễn bằng cách sử dụng dữ liệu nghiên cứu liên kết toàn bộ gen.Phương phápChúng tôi mô tả kết quả từ một nghiên cứu liên kết toàn bộ gen về hen suyễn được thực hiện trên 3855 đối tượng, sử dụng một bảng các 455 089 biến thể đơn nucleotide (SNPs).Kết quảNghiên cứu liên kết toàn bộ gen đã dẫn đến việc ưu tiên 33 biến thể cho theo dõi ngay lập tức trong một nỗ lực tái lập nhiều giai đoạn. Trong số này, một biến thể phổ biến (rs9272346) nằm trong khoảng 1 Kb của HLA‐DQA1 (nhiễm sắc thể 6p21.3) có liên quan đến hen suyễn ở người lớn (P‐giá trị = 2.2E‐08) với bằng chứng nhất quán trong nhóm người lớn và trẻ em đa dạng hơn (P‐giá trị = 1.0E‐04). Hơn nữa, một số gen đã được xác định trong các GWAS hen suyễn trước đây cũng có liên quan đến hen suyễn trong quần thể của chúng tôi.Kết luậnTổng thể, các phát hiện của chúng tôi càng khẳng định thêm vùng HLA‐DQ trong bệnh sinh của hen suyễn. HLA‐DQA1 là thành viên thứ tư của gia đình HLA được phát hiện có liên quan đến hen suyễn, bên cạnh các biến thể đã được xác định trước đó là HLA‐DRA, HLA‐DQB1 và HLA‐DQA2.
