Misoprostol đã được đánh giá trên những đối tượng khỏe mạnh về cả hoạt tính chống tiết và dược lý. Liều lượng sử dụng được xác định ban đầu từ việc kiểm tra sự chịu đựng cấp tính và mãn tính ở những đối tượng khỏe mạnh. Trong khoảng liều đơn từ 50–200 μg, misoprostol ức chế sự tiết acid dạ dày theo cách phụ thuộc liều cả trong trạng thái cơ bản và sau các kích thích như histamin và các bữa ăn thử tiêu chuẩn. Liều 200 μg khác biệt đáng kể so với giả dược như một tác nhân chống tiết. Một nghiên cứu sơ bộ trên sáu đối tượng cho thấy rằng liều 400 μg không tạo ra sự gia tăng đáng kể về hoạt tính so với liều 200 μg. Hơn nữa, các tác dụng phụ như tiêu chảy và cơn co thắt bụng dường như cũng có liên quan đến liều. Hành động chống tiết của misoprostol đạt tối đa một giờ sau khi dùng thuốc và không đáng kể sau 4–5 giờ. Những yếu tố này cho đến nay đã quyết định một chế độ liều lượng 50–200 μg bốn lần một ngày cho misoprostol trong các thử nghiệm lâm sàng chống loét dạ dày. Các phép đo FEV1, dung tích sống và lưu lượng thở ra tối đa cho thấy rằng misoprostol không có hiệu ứng giãn phế quản hay co thắt phế quản đáng kể. Các nghiên cứu về chức năng nội tiết chỉ cho thấy một sự gia tăng nhẹ trong khoảng bình thường của cortisol huyết thanh ở phụ nữ.
