Mô hình xương sọ chuột đã được sử dụng rộng rãi trong nghiên cứu in vivo về tình trạng tiêu xương do hạt, nguyên nhân thường gặp nhất gây ra hiện tượng lỏng vô căn của các khớp nhân tạo. Theo cách truyền thống, mô hình này sử dụng một kỹ thuật phẫu thuật mở, trong đó các hạt polyethylene (PE) được trải trực tiếp lên xương sọ để kích thích tiêu xương. Chúng tôi đã đánh giá một phương pháp điều chỉnh xâm lấn tối thiểu cho mô hình xương sọ bằng cách tiêm trực tiếp các hạt PE dưới da. Các hạt polyethylene (PE) được tiêm dưới da trên xương sọ của những con chuột C57BL6J mười tuần tuổi (nhóm “tiêm”) hoặc được cấy ghép sau khi phẫu thuật lộ xương sọ (nhóm “mở”) (n = 5/nhóm). Đối với mỗi nhóm, năm con chuột khác không nhận hạt và phục vụ như nhóm đối chứng. Tình trạng tiêu xương do hạt được đánh giá hai tuần sau khi thực hiện thủ thuật bằng cách sử dụng hình ảnh vi tính độ phân giải cao microCT. Việc tiêm các hạt polyethylene vào xương sọ dẫn đến giảm 40% ± 1.8% khối lượng xương (BVF) so với nhóm đối chứng. Sử dụng “kỹ thuật phẫu thuật mở”, BVF giảm 16% ± 3.8% so với nhóm đối chứng (p < 0.0001). Tiêm trực tiếp các hạt PE dưới da trên xương sọ chuột tạo ra tình trạng tiêu xương đáng kể hơn. Việc cấy ghép hạt polyethylene qua tiêm ít xâm lấn hơn và dẫn đến tình trạng tiêu xương đáng kể hơn so với kỹ thuật mở. Phương pháp tiêm dưới da này sẽ hữu ích cho các lần tiêm lặp lại các hạt và đánh giá các liệu pháp tiềm năng tại chỗ hoặc toàn thân.
