Acid béo không bão hòa đa n‐3 trong chế độ ăn ảnh hưởng đến sự phát triển của tăng huyết áp thận mạch ở chuột

Hậu quả của việc cung cấp n‐3 PUFA qua chế độ ăn đã được nghiên cứu về sự gia tăng huyết áp (BP) do hẹp động mạch thận trái gây ra ở chuột, được chia thành 3 nhóm (n = 8) được cho ăn trong 8 tuần bằng chế độ ăn bán tinh khiết, bao gồm chế độ ăn đối chứng hoặc chế độ ăn giàu acid béo (acid docosahexaenoic, DHA, hoặc acid eicosapentaenoic, EPA). Việc tiêu thụ PUFA đã gây ra những thay đổi lớn trong hồ sơ acid béo phospholipid ở tim và thận, nhưng không ảnh hưởng đến cân nặng cơ thể, phì đại cơ tim, teo thận trái và phì đại thận phải. Trong vòng 4 tuần, huyết áp tăng từ 120–180 ± 2 mm Hg ở nhóm đối chứng, nhưng chỉ lên đến 165 ± 3 mm Hg ở các nhóm n‐3 PUFA. Sau khi huyết áp được ổn định trong 3 nhóm, chuột được đưa vào một liệu pháp ngắn hạn với liều tăng dần của perindopril. Liều thấp hơn (0.5 mg/kg) làm giảm huyết áp một cách vừa phải chỉ ở nhóm đối chứng. Với các liều cao hơn (1, 5 và 10 mg/kg), huyết áp được điều chỉnh về mức bình thường trong cả 3 nhóm, với biên độ giảm huyết áp lớn hơn ở nhóm đối chứng. Việc tiêu thụ n‐3 PUFA ở mức độ vừa phải có thể giúp ngăn chặn sự phát triển của tăng huyết áp ngoại vi ở chuột thông qua một cơ chế có thể liên quan đến enzyme chuyển đổi angiotensin.
