Tính chất chống ung thư của chiết xuất hydro-alcool của củ dền đỏ và thành phần chính của nó; betanin trên các dòng tế bào ung thư đại trực tràng

Tóm tắt

Ung thư đại trực tràng (CRC) là loại ung thư phổ biến thứ ba trên thế giới. Củ dền đỏ (Beta vulgaris) chứa Betanin là betacyanin chính, sở hữu nhiều hiệu ứng proapoptotic. Nghiên cứu này nhằm mục đích điều tra các hiệu ứng chống ung thư và proapoptotic của chiết xuất hydro-alcool củ dền (BHE) và betanin, trên các dòng tế bào ung thư đại trực tràng. BHE và betanin được sử dụng để điều trị các tế bào ung thư đại trực tràng Caco-2 và HT-29. Các thử nghiệm MTT, nhuộm DAPI và phân tích tế bào FACS đã được sử dụng để xác định nồng độ ức chế tối thiểu (IC50) và đánh giá sự kích thích apoptosis. Các gen được mong muốn được đánh giá bằng phương pháp phản ứng chuỗi polymerase theo thời gian thực (RT-PCR). IC50 cho các dòng tế bào HT-29 và Caco-2 lần lượt là 92 μg/mL, 107 μg/mL cho BHE, và 64 μg/mL, 90 μg/mL cho betanin ở 48 giờ. BHE và betanin ức chế đáng kể sự tăng trưởng của cả hai dòng tế bào ung thư theo thời gian và liều lượng phụ thuộc. Kết quả nhuộm DAPI và phân tích tế bào dòng cho thấy triệu chứng apoptosis đáng kể ở các dòng tế bào ung thư đã được điều trị. Mức độ biểu hiện của các gen proapoptotic (BAD, Caspase-3, Caspase-8, Caspase-9 và Fas-R) trong các tế bào HT-29 và Caco-2 đã được điều trị cao hơn so với các tế bào không được điều trị và tế bào bình thường. Ngược lại, gen chống apoptosis (Bcl-2) đã được điều chỉnh giảm một cách đáng kể. BHE và betanin đã ức chế hiệu quả sự sinh sản tế bào ung thư và kích thích apoptosis thông qua việc điều chỉnh các gen hiệu quả.
