Để điều tra xem một kiểu loại thụ thể cụ thể có thể liên kết với nhiều hệ thống tác động khác nhau không, đã tiến hành biểu hiện các thụ thể muscarinic M2 tái tổ hợp trong các tế bào không có thụ thể nội sinh. Chất kích thích muscarinic là carbachol vừa ức chế adenylyl cyclase vừa kích thích thủy phân phosphoinositide. Việc kích thích sự thủy phân phosphoinositide ít hiệu quả hơn một cách đáng kể và phụ thuộc nhiều hơn vào mức độ thụ thể so với việc ức chế adenylyl cyclase. Cả hai phản ứng đều được trung gian bởi các protein liên kết với guaninnucleotide, như đã được chứng minh bằng việc chúng bị ức chế bởi độc tố ho gà; phản ứng adenylyl cyclase được liên kết hiệu quả hơn thì nhạy cảm hơn rất nhiều.
