Có nhiều chứng cứ cho thấy yếu tố tăng trưởng nội mô mạch máu (VEGF) có liên quan đến việc tạo mạch và sự phát triển của các khối u nguyên phát cũng như hình thành di căn. Sự biểu hiện của yếu tố tăng trưởng nội mô mạch máu phụ thuộc vào các oncogen đã được kích hoạt và các gen ức chế khối u đã bị vô hiệu hóa, cũng như một số yếu tố khác, chẳng hạn như yếu tố tăng trưởng, tình trạng thiếu oxy và các chất thúc đẩy khối u. Sự biểu hiện đáng kể của các thụ thể yếu tố tăng trưởng nội mô mạch máu chủ yếu bị giới hạn ở các mạch máu của khối u. Sự tham gia nguyên nhân của yếu tố sinh mạch này trong tiến trình phát triển của bệnh đã được đánh giá thành công bằng cách sử dụng kháng thể đơn dòng chống lại yếu tố tăng trưởng nội mô mạch máu, đột biến thụ thể loại bỏ phế và các oligonucleotide chống cảm ứng chống lại RNA thông tin của yếu tố tăng trưởng nội mô mạch máu. Do đó, hệ thống tín hiệu VEGF dường như là một mục tiêu thích hợp để ức chế sự sinh mạch khối u và hình thành di căn.
