Khử nitrat là một quá trình có tầm quan trọng môi trường lớn nhưng rất khó nghiên cứu trong các hệ sinh thái trên cạn. Các phương pháp định lượng quá trình này gặp nhiều vấn đề, tính biến động trong hoạt động rất cao, và những thách thức về quy mô theo thời gian và không gian thì rất cực đoan. Các phương pháp hiện có gặp nhiều vấn đề vì nhiều lý do; chúng thay đổi nồng độ chất nền, làm rối loạn môi trường vật lý của quá trình, thiếu độ nhạy hoặc tốn kém rất nhiều thời gian và chi phí. Vấn đề cơ bản nhất là rất khó định lượng sản phẩm cuối chiếm ưu thế (N2) của quá trình khử nitrat do nồng độ nền cao của nó trong khí quyển. Sự biến động không gian và thời gian trong quá trình khử nitrat rất cao vì quá trình này chịu sự chi phối của nhiều yếu tố (oxy, nitrat, carbon, pH, độ mặn, nhiệt độ, v.v.) mà mỗi yếu tố đều thay đổi theo thời gian và không gian. Một thách thức đặc biệt là các vùng nhỏ (trung tâm nóng) và thời gian ngắn (thời điểm nóng) thường chiếm tỷ lệ phần trăm cao trong hoạt động dòng khí N. Những hiện tượng này rất khó để tính toán trong các nỗ lực đo lường, mô hình hóa và quy mô. Nhu cầu cho việc quy mô là do sự cần thiết thông tin về quá trình vi mô này ở quy mô hệ sinh thái, cảnh quan và khu vực nơi có lo ngại về tác động của nitơ đến độ phì nhiêu của đất, chất lượng nước và chất lượng không khí.
