Sansoninto như một loại thuốc điều trị dựa trên bằng chứng cho hành vi giống trầm cảm

Phương pháp sàng lọc trong ống nghiệm với tế bào Neuro2a nuôi cấy để kiểm tra sự hoạt hóa (phosphorylation) của kinase ngoại bào điều chỉnh tín hiệu (ERK) 1/2 và thúc đẩy sự phát triển của nhánh thần kinh đã cho thấy rằng chiết xuất từ 5 công thức Kampo (truyền thống Nhật Bản) có tiềm năng như thuốc điều trị các bất thường về hành vi. Bởi vì chiết xuất sansoninto (SAT) thể hiện tác dụng mạnh hơn so với các ứng cử viên khác được thử nghiệm, chúng tôi đã điều tra xem việc sử dụng miệng của nó có cải thiện được các bệnh lý của một số mô hình chuột mắc chứng suy giảm hành vi hay không. Kết quả thu được cho thấy chiết xuất SAT có tác dụng chống trầm cảm trong bài kiểm tra bơi cưỡng bức, điều này có thể được điều tiết bởi sự tăng cường biểu hiện của yếu tố dinh dưỡng thần kinh có nguồn gốc từ não (BDNF) trong hồi hải mã. Nó cũng có thể được điều tiết bởi sự phosphory hóa tăng cường của protein liên kết yếu tố phản ứng cAMP (CREB) thông qua chuỗi tín hiệu kinase được kích hoạt bởi mitogen (MAPK) và chuỗi tín hiệu kinase protein phụ thuộc vào Ca2+/calmodulin (CaMK II), một chuỗi tín hiệu hạ nguồn của thụ thể N-methyl-d-aspartate (NMDA).
