Carbohydrate là nguồn năng lượng tiết kiệm nhất trong thức ăn cho cá, nhưng hầu hết các loài cá có khả năng sử dụng carbohydrate hạn chế. Có báo cáo rằng phosphoenolpyruvate carboxykinase 1 (pck1) tham gia vào quá trình chuyển hóa carbohydrate, chuyển hóa lipid và các quá trình trao đổi chất khác. Tuy nhiên, vẫn thiếu các bằng chứng trực tiếp để hiểu rõ về mối quan hệ giữa pck1 và chuyển hóa glucose cùng lipid. Ở đây, chúng tôi đã tạo ra một dòng cá bảy màu bị knockout pck1 thông qua hệ thống CRISPR/cas9, và cung cấp chế độ ăn giàu carbohydrate cho cá từ 60 ngày sau thụ tinh (dpf) trong 8 tuần. Chúng tôi phát hiện rằng cá bảy màu thiếu pck1 có nồng độ glucose trong huyết tương giảm, mức độ mRNA của các gen liên quan đến đường phân (gck, pfk, pk) tăng cao, và các mức độ phiên mã của các gen gluconeogenic (pck1, fbp1a) trong gan giảm. Chúng tôi cũng nhận thấy nồng độ triglyceride, cholesterol toàn phần và sự tích tụ lipid giảm ở cá pck1−/−, cùng với việc giảm biểu hiện của các gen liên quan đến lipolysis (acaca) và lipogenesis (cpt1).
