Cấu trúc gập của protein được bảo tồn hơn so với chuỗi amino acid của nó và có mối liên hệ chặt chẽ với chức năng sinh học, do đó việc tính toán sự tương đồng của các cấu trúc protein là một vấn đề cơ bản trong sinh học cấu trúc và đóng vai trò then chốt trong phân loại cấu trúc protein, suy diễn chức năng gập và dự đoán cấu trúc protein. Trong những năm gần đây, đã có nhiều tiến bộ lớn trong lĩnh vực này và nhiều phương pháp để xem xét sự tương đồng cấu trúc đã được đề xuất, bao gồm các phương pháp so sánh cấu trúc protein, truy xuất cấu trúc protein từ cơ sở dữ liệu và so sánh vị trí liên kết ligand. Hầu hết các phương pháp này có thể truy cập trên Mạng toàn cầu, nhưng việc đánh giá tất cả các phương pháp vẫn là một vấn đề khó khăn. Bài báo này tóm tắt một số phương pháp phổ biến và những phương pháp mới nhất cho sự tương đồng cấu trúc, bao gồm căn chỉnh cấu trúc, truy xuất cấu trúc protein và căn chỉnh vị trí liên kết ligand.
