Tác động bảo vệ của polyphenol trà đối với hàng rào máu-não

Nghiên cứu này nhằm điều tra tác động bảo vệ của các polyphenol trong trà đối với hàng rào máu-não (BBB) của chuột cống trong tình trạng tổn thương thiếu oxy/toàn bộ não (GCIR). Chuột Sprague Dawley đã trải qua phẫu thuật tắc mạch bốn mạch để xây dựng mô hình GCIR. Nửa giờ trước khi tắc hoàn toàn, chúng được điều trị bằng polyphenol trà (TP) (6,4%; 100 hoặc 200 mg/kg) qua tiêm tĩnh mạch vào đuôi. 24 giờ sau khi hồi phục tưới máu, độ thẩm thấu của BBB được đánh giá thông qua việc đo hàm lượng nước trong não (BWC) và lượng thuốc nhuộm Evan’s blue còn lại trong mô não. Ngoài ra, biểu hiện protein MMP-9 và collagen IV trong mô não cũng được phát hiện bằng phương pháp hóa miễn dịch. Phân tích thống kê được thực hiện bằng ANOVA và SNK-q. Ý nghĩa thống kê được xem xét khi P<0,05. So với nhóm không điều trị, chuột được điều trị bằng TP có BWC đáng kể giảm (P<0,05), lượng thuốc nhuộm Evan’s blue còn lại trong mô não giảm (P<0,05), mức MMP-9 giảm xuống (P<0,05) và biểu hiện collagen IV tăng lên trong mô não (P<0,05). Từ những phát hiện này, có thể kết luận rằng TP có thể làm giảm sự phân hủy collagen IV do MMP-9 gây ra bởi GCIR nhằm bảo vệ BBB.
