Mặc dù được sử dụng rộng rãi như một phương pháp bảo quản, việc đông lạnh vẫn gặp khó khăn trong việc bảo quản tảo vi sinh do mức sống sót thấp và tính đặc hiệu của chủng. Trong nghiên cứu này, một phương pháp đông lạnh cho Ettlia sp. YC001 đã được tối ưu hóa sử dụng các chiến lược sau. Việc thích ứng lạnh ở nhiệt độ 4 °C trong 24 giờ trước quy trình đông lạnh đã làm tăng sự biểu hiện của con đường phản ứng lạnh và các protein sốc nhiệt, giúp ổn định các protein chống lại sự căng thẳng do lạnh và ngăn ngừa mất nước nhanh chóng. Ba thông số độc lập trong quá trình đông lạnh, bao gồm tốc độ làm lạnh, nhiệt độ ngâm, và nhiệt độ rã đông, đã được tối ưu hóa với nhiều chất bảo vệ đông lạnh khác nhau bằng cách sử dụng phương pháp mặt phẳng phản ứng (RSM). Cuối cùng, tổn thương tế bào đã được giảm thiểu khi sử dụng quy trình làm lạnh có kiểm soát tối ưu với tủ đông lập trình chính xác. Khi áp dụng các điều kiện tối ưu, tỷ lệ sống của chủng tảo đã được đông lạnh YC001 > 90% sau 1 năm. Được cho là phương pháp đề xuất cũng có thể áp dụng cho các chủng tảo vi sinh thương mại khác.
