Xác định và xác thực QTL cho khả năng chịu lạnh ở giai đoạn cây con trong lúa từ giống indica Habiganj Boro VI (Hbj.BVI)

Hiện tượng vàng lá, sự phát triển yếu và chết cây con do stress nhiệt độ thấp là một vấn đề phổ biến ở nhiều quốc gia châu Á trong việc trồng lúa mùa đông. Cải thiện các giống lúa thông qua chọn lọc hỗ trợ bởi dấu phân tử QTL cho khả năng chịu lạnh ở giai đoạn cây con từ nguồn giống bản địa là chiến lược hiệu quả và bền vững nhất để giải quyết vấn đề này. Một nghiên cứu đã được thực hiện để xác định vị trí các QTL từ 151 dòng F2:3 của một phép lai giữa giống lúa nhạy cảm với lạnh BR1 và một giống lúa indica truyền thống thích ứng địa phương, Hbj.BVI. Tổng cộng có sáu QTL đáng kể đã được xác định cho hai chỉ số khả năng chịu lạnh - sự đổi màu lá do lạnh và tỷ lệ sống sót sau thời gian phục hồi bảy ngày trên nhiễm sắc thể 6, 8, 11 và 12. Trong số các QTL này, qCTSL-8-1 và qCTSS-8-1 được đồng vị trí trong khoảng RM7027–RM339 trên nhiễm sắc thể 8, và qCTSL-12-1 và qCTSS-12-1 trong khoảng RM247–RM2529 trên nhiễm sắc thể 12 hiển thị mức đóng góp lần lượt là 12,78% và 14,96% vào biến động kiểu hình tổng thể đối với khả năng chịu lạnh. Việc xác thực hiệu ứng của QTL trong quần thể BC1F3 từ phép lai giữa BRRI dhan28 nhạy cảm với lạnh và Hbj.BVI cho thấy tác động vượt trội của qCTSL-12-1 đối với khả năng chịu lạnh ở giai đoạn cây con và tác động này mạnh hơn khi một hoặc nhiều QTL khác phân ly cùng với nó. Những kết quả này cho thấy rằng các QTL được xác định trong nghiên cứu này là ổn định và hiệu quả trên các nền tảng di truyền khác, điều này khẳng định việc sử dụng các QTL này cho các nghiên cứu tiếp theo nhằm phát triển giống có khả năng chịu lạnh ở giai đoạn cây con.
