Die Effektivität der Expression von Fremdgenen kann nicht nur auf der Ebene der Transkription, sondern auch auf posttranskriptionellem Niveau erhöht werden. Durch Nutzung viraler genetischer Systeme, die sich unabhängig von ihrem Wirt replizieren, erscheint es möglich, die Genproduktmenge um Größenordnungen gegenüber dem bisher experimentell erreichbaren Niveau zu steigern. Der Grundgedanke besteht darin, einerseits virale Geninformation für eine funktionell aktive RNA-abhängige RNA-Polymerase stabil zu transferieren und zu exprimieren und andererseits die mRNA, die für das interessierende Genprodukt kodiert, mit den Erkennungssequenzen für die Virusreplikase auszustatten, so daß es zu der gewünschten mRNA-Amplifikation kommt.
