Béo phì, tăng huyết áp và tiền đái tháo đường đóng góp lớn vào bệnh động mạch vành, suy tim và các biến cố mạch máu, và là nguyên nhân chính gây tử vong và bệnh tật trong các xã hội phát triển. Nhạy cảm với muối làm trầm trọng thêm rối loạn chức năng nội mô. Trong nghiên cứu này, chúng tôi đã điều tra tác động của việc kích hoạt thụ thể peptide giống glucagon-1 (GLP-1) mạn tính lên vi tuần hoàn vành và tái tạo tim trong chuột Zucker trên chế độ ăn nhiều muối (6% NaCl).

Phương pháp

Các con chuột Zucker gầy (+/+) và béo phì (fa/fa) 8 tuần tuổi được điều trị bằng dung dịch hoặc liraglutide (LIRA) (0,1 mg/kg/ngày, tiêm dưới da) trong 8 tuần. Huyết áp tâm thu (SBP) được đo bằng phương pháp vòng đuôi ở chuột tỉnh. Chức năng cơ tim được đánh giá bằng siêu âm tim. Kỹ thuật vi mạch tương phản đồng bộ được sử dụng để điều tra chức năng mạch vành (mạch có đường kính trong 50–350 µm) in vivo trên chuột đã gây mê. Mức độ biểu hiện gen và protein của các yếu tố vasoactive, dấu hiệu viêm, căng thẳng oxy hóa và tái tạo được xác định bằng PCR thời gian thực và phương pháp Western blot.
