Chúng tôi trước đây đã báo cáo rằng các tế bào dendritic tủy nhân tạo (pDC) trong các khối u vú nguyên phát có mối tương quan với tiên lượng không thuận lợi cho bệnh nhân. Nghiên cứu hiện tại được thiết kế để giải mã cơ sở cơ chế cho tác động bất lợi của pDC đối với kết quả lâm sàng. Sử dụng các mẫu sinh thiết khối u vú người tươi (N = 60 bệnh nhân), chúng tôi quan sát thấy qua phân tích tế bào học bằng dòng đa tham số, tỷ lệ các pDC liên kết với khối u (TApDC) gia tăng trong các khối u vú hung hãn, tức là những khối u có chỉ số phân chia cao và được gọi là các khối u vú ba âm tính (TNBT). Hơn nữa, TApDC thể hiện kiểu hình một phần đã được kích hoạt và sản xuất lượng IFN-α rất thấp sau khi kích hoạt thụ thể như toll trong ống nghiệm so với pDC máu của bệnh nhân. Trong các khối u vú, TApDC đã đồng địa điểm và có mối tương quan mạnh với các tế bào T điều hòa TA (TATreg), đặc biệt trong TNBT. Điều quan trọng nhất, sự ức chế chọn lọc sản xuất IFN-α đã trao cho TApDC khả năng đặc biệt duy trì sự mở rộng của Treg FoxP3+, một khả năng đã được phục hồi bằng cách thêm IFN-α ngoại sinh. Những phát hiện này cho thấy rằng TApDC thiếu IFN-α tích tụ trong các khối u hung hãn có liên quan đến sự mở rộng của TATreg in vivo, góp phần vào sự dung nạp miễn dịch của khối u và kết quả lâm sàng kém.
