Liệu pháp hóa-xạ nhiệt có thể cải thiện hiệu quả điều trị của hóa trị liệu đơn giản với sự hỗ trợ từ nhiệt độc tế bào sinh ra bởi liệu pháp quang nhiệt. Trong bài báo này, một nền tảng nano mới dựa trên silica rỗng mesoporous (HMSN) đã được xây dựng, có khả năng đạt được giải phóng đáp ứng đa kích thích thông qua tương tác hóa-xạ nhiệt. Cypate, một tác nhân quang nhiệt NIR, được liên kết với bề mặt ngoài của HMSN bằng các liên kết disulfide. Nhờ vào tính chất không tan, Cypate đã impart lipophilicity cho bề mặt của HMSN, do đó thúc đẩy quá trình bao bọc mạnh mẽ của lớp phospholipid phức tạp thông qua tương tác kỵ nước. Lớp phospholipid đã cản trở sự giải phóng sớm của thuốc mẫu doxorubicin (DOX), kéo dài thời gian tuần hoàn trong cơ thể sống, và cải thiện khả năng tương thích sinh học cũng như độ ổn định trong môi trường sinh lý. Dưới các điều kiện gluthathione, axit và bức xạ NIR, sự giải phóng DOX đã được gia tốc đáng kể, cho thấy DOX/HMSN-CyL thể hiện phản ứng đa kích thích redox/pH/NIR đối với việc giải phóng thuốc. Các thí nghiệm độc tính tế bào đã chứng minh rằng liệu pháp kết hợp đã gây ra mức độ tiêu diệt tế bào tối đa. Hơn nữa, chỉ số kết hợp của DOX/HMSN-CyL là 0.26, cho thấy liệu pháp synergistic hóa-xạ nhiệt. Các thí nghiệm spheroid khối u đa tế bào cũng được tiến hành, và xu hướng tương tự đã được thể hiện. Tóm lại, nền tảng nano mới này có triển vọng ứng dụng rộng rãi như một hệ thống phân phối thuốc cho liệu pháp kết hợp.
