Characterization of Citric Acid Biosynthesis by Fungi Aspergillus niger for Surface and Submerged Cultivation

Citric acid is an organic acid found in the highest concentration in a number of plants and widely used in many branches of industry. In addition, it is a one of the metabolites synthesizing by many microorganisms, especially fungi. The aim of this work was the comparing of citric acid biosynthesis under various conditions of Aspergillus niger cultivation on an inulin-containing extract obtained from Jerusalem artichoke plant tubers. By spectrophotometric method, 7% inulin was determined in the extract. The cultivation of fungi was carried out during 6 days in two ways: in an incubator for surface cultivation and on a shaker for submerged cultivation. The content of citric acid was determined by High Performance Liquid Chromatography. The results shown, that the maximum amount of citric acid was observed in case of submerged cultivation on the fourth day of growth. The yield of citric acid was 80.5% for submerged cultivation and 61.3% for surface cultivation. As well as, the possibility of using extract of tubers as effective raw material was determined. Thus, it can be seen that the studied strain of Aspergillus niger has a good potential for use as a strain-producer of citric acid using Jerusalem artichoke tuber extract as a growth medium.
