Phân tích DNA methyl hóa trong các mô và giai đoạn phát triển của ngọc trai Pinctada fucata

Trong nghiên cứu này, chúng tôi đã phát triển một kỹ thuật đa hình khuếch đại nhạy cảm với methyl hóa để điều tra hồ sơ methyl hóa DNA trong các mô khác nhau và trong các giai đoạn phát triển sớm của ngọc trai Pinctada fucata (P. fucata). Mức độ methyl hóa trong cơ, tuyến tiêu hóa, chân trục, tim và mang dao động từ 11,71 đến 14,71%, và sự khác biệt đáng kể (P < 0,05) giữa các mức độ methyl hóa trong các mô khác nhau đã được ghi nhận. Mức độ methyl hóa DNA của tinh trùng, tế bào trứng, phôi hai tế bào, phôi morula, ấu trùng trochophore và ấu trùng hình chữ D lần lượt là 13,51, 11,80, 12,14, 12,60, 14,65 và 13,18%. Các giai đoạn phát triển của phôi hai tế bào, phôi morula, ấu trùng trochophore và ấu trùng hình chữ D cho thấy số lượng các locus trạng thái methyl hóa DNA giống nhau trong trứng cao hơn so với trong tinh trùng. Có khả năng rằng các mẫu methyl hóa DNA của thế hệ con chủ yếu bị ảnh hưởng bởi trứng, trong khi tinh trùng có thể ngày càng quan trọng trong quá trình phát triển phôi sớm. Những khác biệt quan sát được trong các mức độ methyl hóa ở các mô và giai đoạn phát triển của P. fucata cho thấy rằng methyl hóa DNA có thể hoạt động như một yếu tố điều chỉnh epigenetic trong quá trình phân hóa mô, tăng trưởng cá nhân và phát triển.
