A simple loop-mediated isothermal amplification assay to detect Phytophthoracolocasiae in infected taro plants

In Japan, Phytophthoracolocasiae causes leaf blight of taro, resulting in huge losses. To effectively diagnose this pathogen in the field, we developed rapid and reliable methods based on loop-mediated isothermal amplification (LAMP) using LAMP primers designed from the Ypt1 gene that are highly specific with a sensitivity of 100 fg. Among these methods, we concluded that the plant-LAMP method with a heat treatment is simple and dependable for detecting P.Colocasiae in taro cultivation fields and that the plant culture-LAMP is a simple and reliable for identifying P.Colocasiae that has been cultured on media for extended periods.
