Phương pháp RT-QPCR mới, có độ chọn lọc cao để định lượng MSRV sử dụng PNA clamping syncytin-1 (ERVWE1)

HERV-W là một gia đình đa lộc của virus retrovirus nội sinh ở người (HERVs) đã được phát hiện là đóng một vai trò quan trọng trong sinh lý và bệnh lý của con người. Hai thành viên đặc biệt của gia đình HERV-W là những đối tượng rất được quan tâm: ERVWE1 (mã hóa syncytin-1, một glycoprotein thiết yếu trong việc hình thành nhau thai) và MSRV (virus retrovirus liên quan đến bệnh đa xơ cứng, được cho là có vai trò quan trọng trong bệnh lý của con người do sự gia tăng biểu hiện của nó trong mô não và tế bào máu lấy từ bệnh nhân mắc bệnh đa xơ cứng (MS)). Cả hai mRNA của ERVWE1 và MSRV có mức độ tương đồng cao và do đó một phương pháp cho phép định lượng sự biểu hiện của MSRV một cách độc quyền trong các mẫu lâm sàng sẽ là mong muốn. Chúng tôi đã phát triển một kỹ thuật phản ứng chuỗi polymerase định lượng (QPCR) để phát hiện và định lượng virus retrovirus liên quan đến bệnh đa xơ cứng. Phép thử sử dụng probe oligonucleotide đánh dấu huỳnh quang, tương thích với đoạn gen env của MSRV và một probe acid nucleic peptide (PNA), hoàn toàn tương thích với đoạn trình tự ERVWE1 mà chặn hiệu quả hoạt động của polymerase trên các mẫu ERVWE1. Phân tử PNA, nếu được sử dụng song song với probe phân hủy trong phân tích QPCR, sẽ tạo điều kiện thuận lợi cho hiệu quả phát hiện của MSRV ngay cả khi ERVWE1 có mặt nhiều hơn đáng kể so với MSRV trong mẫu được phân tích. Chúng tôi đã đạt được một phạm vi rộng và có thể đo được từ 1 × 10e2 đến 1 × 10e8 bản sao/phản ứng; tính tuyến tính của kỹ thuật được duy trì ngay cả ở mức MSRV thấp là 1% so với ERVWE1. Sử dụng phương pháp mới phát triển của chúng tôi, chúng tôi xác nhận rằng sự biểu hiện của MSRV diễn ra trong các tế bào thần kinh đệm của người bình thường và trong tế bào nội mô tĩnh mạch rốn người trong ống nghiệm. Chúng tôi cũng phát hiện rằng sự kích thích của bạch cầu đơn nhân người không ảnh hưởng đến biểu hiện cụ thể của MSRV nhưng đã gây ra những thay đổi ở mức độ mRNA của các mẫu HERV-W khác nhau.
