Effect of Co-incubation with Mesenchymal Stromal Cells in Cultural Medium on Structure and Mechanical Properties of Polylactide-Based Scaffolds

In present work, the influence of cultural medium on structure and properties of polymer-based scaffolds was studied. Polylactide (PLA) was used as the polymer matrix and micro- and nanoparticles of hydroxyapatite were used as the filler. Extrusion was used to obtain the composites. Porous scaffolds were 3D printed. Cultural medium RPMI-1640 was used as the medium for incubation. Compression tests were used to study mechanical properties before and after incubation. Differential scanning calorimetry was used to study influence of hydrolysis on thermal properties of porous scaffolds. Scaffolds were colonized by mesenchymal stromal cells (MSCs). Microstructure was studied by optical microscopy after colonization. Decrease of mechanical properties was observed after incubation. In vitro test showed cytoconductivity of porous scaffolds.
