Differences between the structures of poliovirus and sindbis virus infectious ribonucleic acids

The presence of spermine during heating and quick cooling of the infectious RNA of Sindbis virus increased the survival 11-fold; a four-fold increase was obtained with diaminohexane; no protection was afforded by amines of shortest chain length. None of the amines tested protected poliovirus RNA against heat. The absorbance-temperature profiles were obtained for RNA's extracted from the purified viruses. A sharp hyperchromic effect was shown by Sindbis RNA but not by polio RNA. The increase in optical density was 32% and the transition midpoint 65.5° C. These figures are higher than those published for other single stranded RNA viruses and contrast with the fact that Sindbis virus has not the highest G + C content. The infectivity of Sindbis RNA was less susceptible to the action of pancreatic ribonuclease than was the infectivity of polio RNA. These facts confirm previous findings indicating that Sindbis RNA, in contrast to polio RNA, has a secondary structure, and that this structure is necessary for the infectivity.
