Tác động của việc bổ sung gemcitabine vào điều trị hóa trị xạ 5-fluorouracil (5-FU) đối với tỷ lệ sống sót tổng thể (OS) trong 5 năm ở bệnh nhân đã cắt bỏ adenocarcinoma tuyến tụy được trình bày với những kết quả cập nhật từ một thử nghiệm pha III. Sau khi cắt bỏ adenocarcinoma tuyến tụy, bệnh nhân được phân ngẫu nhiên vào nhóm dùng 5-FU trước và sau hóa trị xạ so với nhóm dùng gemcitabine trước và sau hóa trị xạ. 5-FU được cung cấp liên tục với liều 250 mg/m2/ngày và gemcitabine được cung cấp với liều 1000 mg/m2 hàng tuần. Cả hai loại thuốc đều được dùng trong 3 tuần trước và 12 tuần sau khi hóa trị xạ. Hóa trị xạ được cung cấp với liều 50.4 Gy kết hợp với 5-FU được sử dụng liên tục. Điểm chính được quan sát là tỷ lệ sống cho tất cả bệnh nhân và cho bệnh nhân có khối u ở đầu tụy. Có 451 bệnh nhân đủ điều kiện tham gia. Phân tích đơn biến không cho thấy sự khác biệt về tỷ lệ sống. Bệnh nhân có khối u ở đầu tụy (n = 388) có thời gian sống trung bình và tỷ lệ sống 5 năm lần lượt là 20.5 tháng và 22% với gemcitabine so với 17.1 tháng và 18% với 5-FU.
