Tế bào γδ T có thể đóng vai trò quan trọng trong phòng thủ của cơ thể, và những bằng chứng gần đây đã chỉ ra rằng chúng rất quan trọng trong việc điều tiết phản ứng miễn dịch sau khi gặp phải tác nhân gây bệnh. Dữ liệu cũng đang tích lũy cho thấy chúng đóng vai trò trong tự miễn dịch và điều hòa miễn dịch. Bằng chứng cho thấy tế bào γδ T trung gian những hiệu ứng này thông qua việc sản xuất cytokine, thông qua những tác động trực tiếp lên các tế bào của phản ứng miễn dịch thích ứng, hoặc thông qua các tương tác với các thành phần hoặc rào cản miễn dịch bẩm sinh.
