Here, in a consecutive series of 295 early breast cancer patients, we show that both overall survival and distant metastasis-free survival are markedly diminished in patients whose tumors expressed this wound-response signature compared to tumors that did not express this signature. A gene expression centroid of the wound-response signature provides a basis for prospectively assigning a prognostic score that can be scaled to suit different clinical purposes. Overexpression of h-gas1 is able to block cell\nproliferation in the A549 lung carcinoma and the T24 bladder carcinoma cell\nlines. No effect was observed when h-gas1 was introduced into the osteosarcoma\ncell line SAOS-2 and into the adenovirus-type-5 transformed cell line 293. This\nfinding is related to our previous evidence that simian virus 40-transformed NIH\n3T3 cells are also refractory to murine gas1 overexpression, suggesting that the\nretinoblastoma and\u002For p53 gene products have an active role in mediating the\ngrowth-suppressing effect of gas1. We also show that h-gas1 is on chromosome\n9q21.3-22.1, in a region considered to be a fragile site. sensing is essential for homeostasis in all aerobic organisms, but its mechanism is poorly understood. Data suggest that a phagocytic-like NAD(P)H oxidase producing reactive oxygen species serves as a primary sensor for oxygen. We have characterized a source of superoxide anions in the kidney that we refer to as a renal NAD(P)H oxidase or Renox. Renox is homologous to gp91\u003Cjats:sup>phox\u003C\u002Fjats:sup>(91-kDa subunit of the phagocyte oxidase), the electron-transporting subunit of phagocytic NADPH oxidase, and contains all of the structural motifs considered essential for binding of heme, flavin, and nucleotide.\u003Cjats:italic>In situ\u003C\u002Fjats:italic>RNA hybridization revealed that renox is highly expressed at the site of erythropoietin production in the renal cortex, showing the greatest accumulation of renox mRNA in proximal convoluted tubule epithelial cells. NIH 3T3 fibroblasts overexpressing transfected Renox show increased production of superoxide and develop signs of cellular senescence. DNA fragmentation is mediated by a\n caspase-activated DNA fragmentation factor (DFF)40. Expression and\n folding of DFF40 require the presence of DFF45, which also acts as a\n nuclease inhibitor before DFF40 activation by execution caspases. The\n N-terminal domains (NTDs) of both proteins are homologous, and their\n interaction plays a key role in the proper functioning of this\n two-component system. Here we report that the NTD of DFF45 alone is\n unstructured in solution, and its folding is induced upon binding to\n DFF40 NTD. Peptidoglycans (PGNs) are major components of bacterial cell walls that possess immunity-stimulating activities in metazoans and plants. Here we show that PGN sensing and immunity to bacterial infection in\n            \u003Cjats:italic>Arabidopsis thaliana\u003C\u002Fjats:italic>\n            requires three lysin-motif (LysM) domain proteins. LYM1 and LYM3 are plasma membrane proteins that physically interact with PGNs and mediate\n            \u003Cjats:italic>Arabidopsis\u003C\u002Fjats:italic>\n            sensitivity to structurally different PGNs from Gram-negative and Gram-positive bacteria.\n            \u003Cjats:italic>lym1\u003C\u002Fjats:italic>\n            and\n            \u003Cjats:italic>lym3\u003C\u002Fjats:italic>\n            mutants lack PGN-induced changes in transcriptome activity patterns, but respond to fungus-derived chitin, a pattern structurally related to PGNs, in a wild-type manner. Biodiversity promotes forest ecosystem performance and resistance to insect pests and diseases, but whether or not diverse forests are also better adapted to deal with drought stress remains unknown. Within our study network of 160 forest stands across Europe, we found that mixed species forests are less exposed to drought stress in some regions only. Therefore, managing forest ecosystems for high tree species diversity does not necessarily assure improved resistance to the more severe and frequent drought events predicted for the A major debilitating phenotype in affected females is frequent apneas, and heterozygous\u003Cjats:italic>Mecp2\u003C\u002Fjats:italic>-deficient female mice mimic the human respiratory disorder. GABA defects have been demonstrated in the brainstem of Mecp2-deficient mice. Here, using an intact respiratory network, we show that apnea in RTT mice is characterized by excessive excitatory activity in expiratory cranial and spinal nerves. Augmenting GABA markedly improves the respiratory phenotype. In addition, a serotonin 1a receptor agonist that depresses expiratory neuron activity also reduces apnea, corrects the irregular breathing pattern, and prolongs survival in MeCP2 null males. Combining a GABA reuptake blocker with a serotonin 1a agonist in heterozygous females completely corrects their respiratory defects. Differential screening for cell volume sensitive transcripts in a human hepatoma cell line revealed a gene for a putative serine\u002Fthreonine kinase, h-sgk, which has 98% sequence identity to a serum- and glucocorticoid regulated kinase, sgk, cloned from a rat mammary tumor cell line. h-sgk transcript levels were strongly altered during anisotonic and isotonic cell volume changes. Within 30 min h-sgk RNA was, independent of\n            \u003Cjats:italic>de novo\u003C\u002Fjats:italic>\n            protein synthesis, induced upon cell shrinkage and, due to a complete stop in h-sgk transcription, reduced upon cell swelling. Comparable changes of sgk transcript levels were observed in a renal epithelial cell line. h-sgk mRNA was detected in all human tissues tested, with the highest levels in pancreas, liver, and heart. of the p53 gene is one of the most frequent genetic changes found in human cancers. Recent experiments indicated that p53 might contain a transcription-activating domain, which functions when directed to a promoter. This study shows that wild-type p53 suppresses transcription of the retinoblastoma (Rb) gene. From deletion and mutagenesis experiments, a cis-acting element (GGAAGTGA) susceptible to regulation by p53 was mapped within the Rb promoter. This element overlaps the basal transcription unit of the Rb promoter, suggesting that p53 suppresses Rb transcription through inhibition of the basal promoter activity. The N-terminal acidic and C-terminal basic domains of p53 were both required for this suppression.
