A major objective of the current study was to compare the conidial photosensitization of two fungal species (\u003Cjats:italic>Metarhizium anisopliae\u003C\u002Fjats:italic> and \u003Cjats:italic>Aspergillus nidulans\u003C\u002Fjats:italic>) with methylene blue (MB) and toluidine blue (TBO) under different incubation and light conditions. Parameters examined were media, photosensitizer (PS) concentration and light source. PDT with MB and TBO resulted in an incomplete inactivation of the conidia of both fungal species. Conidial inactivation reached up to 99.7%, but none of the treatments was sufficient to achieve a 100% fungicidal effect using either MB or TBO. PDT delayed the germination of the surviving conidia. Washing the conidia to remove unbound PS before light exposure drastically reduced the photosensitization of \u003Cjats:italic>A. nidulans\u003C\u002Fjats:italic>. The reduction was much smaller in \u003Cjats:italic>M. anisopliae\u003C\u002Fjats:italic> conidia, indicating that the conidia of the two species interact differently with MB and TBO. Conidia of green and yellow \u003Cjats:italic>M. anisopliae\u003C\u002Fjats:italic> mutants were less affected by PDT than mutants with white and violet conidia. The thermal sensitivity of NBD is primarily a consequence of rapid, heat‐induced electronic changes, which increase the observed fluorescence decay rate. As a result, fluorescence intensity and lifetime variations of membrane‐bound NBD‐conjugated phospholipids and fatty acids can be directly correlated with cellular temperature. In contrast, laurdan fluorescence undergoes a dramatic temperature‐dependent Stokes shift as the membrane undergoes a gel‐to‐liquid‐crystalline phase transition. This facilitates the use of fluorescence spectra to record the indirect effect of microenvironmental changes, which occur during bilayer heating. Microscope and suspension measurements of cells and phospholipid vesicles are compared for both probes using steady‐state and fluorescence lifetime (suspension only) data. Our results show that NBD fluorescence lifetime recordings can provide reasonable temperature resolution (approximately 2°C) over a broad temperature range. Laurdan's microenvironmental sensitivity permits better temperature resolution (0.1‐1°C) at the expense of a more limited dynamic range that is determined solely by bilayer properties. The temperature sensitivity of NBD is based on rapid intramolecular rotations and vibrations, while laurdan relies on a slower, multistep mechanism involving bilayer rearrangement, water penetration and intermolecular processes. The firefly luciferase activity of a series of deletion and site‐directed mutants in the amino‐terminal region was investigated \u003Cjats:italic>in vitro\u003C\u002Fjats:italic> and \u003Cjats:italic>in vivo.\u003C\u002Fjats:italic> The mutant luciferases were produced either by \u003Cjats:italic>in vitro\u003C\u002Fjats:italic> transcription and translation or by expressing the cDNA encoding firefly luciferase in the yeast \u003Cjats:italic>Saccha‐romyces cerevisiae\u003C\u002Fjats:italic> as a fusion protein to the galactose DNA binding domain protein. Deleting the N‐terminal amino acid residues from 3 to 10 dramatically reduced the luciferase activity to less than 1% of the wild‐type activity. A marked decrease in the activity (to 5%) was also observed with the Lys8Glu mutant. The Glyl 0Arg and ProllAla mutants showed20–30% activity compared to the wild type. On the other hand, mutant Asn6Lys retained the wild‐type activity level. Randomizing3–11 N‐terminal amino acids also showed a marked decrease in activity (1%). The mutant luciferases with extremely low levels of enzymatic activity were thermally unstable. These mutational and stability data correlate with the crystal structure of firefly luciferase in which specific amino acids in the N‐terminal region form hydrogen bonds to the amino acids in the neighboring p‐stranded sheet. Because the N‐terminal region is not part of the active site, the present results suggest that the highly conserved N‐terminal amino acid sequences of the firefly luciferase are important for The extent of degradation was monitored by measurement of malanaldehyde. This photodegradative process, which we believe is triggered by light catalyzed generation of superoxide and its subsequent transformation to other potent oxidants. was observed to be thwarted substantially if the medium of organ culture was fortified with 10\u003Cjats:sup>−3\u003C\u002Fjats:sup> and 10\u003Cjats:sup>7\u003C\u002Fjats:sup>\u003Cjats:italic>M\u003C\u002Fjats:italic> vitamin E (α‐tochopherol). These studies suggest that vitamin E may be metabolically beneficial by protecting light exposed tissues, such as those in the eye against photoperoxidativc damage concomitant to light‐catalyzed generation of oxygen‐free radicals. The findings appear relevant to age‐associated pathogenesisof cataracts and their possible attenuation.
