Mat structure did not vary with temperature. All mats possessed highly ordered layers of airspaces at both the macroscopic and microscopic level, and these appear to be an adaptation to a free-floating growth habit. Upper mat layers supported biomass with elevated carotenoid:chlorophyll a ratios and an as yet uncharacterized waxy layer on the dorsal surface. Microscopic examination revealed mats comprised a single Fischerella morphotype, with abundant heterocysts throughout mats at all temperatures. Molecular analysis of mat community structure only partly matched morphological identification. All samples supported greater 16S rDNA-defined diversity than morphology suggested, with a progressive loss in the number of genotypes with increasing temperature. Fischerella-like sequences were recovered from mats occurring at all temperatures, but some mats also yielded Oscillatoria-like sequences, although corresponding phenotypes were not observed. enrichment culture from the soil of a forest near Manali, Himachal Pradesh, India, and was identified as Nocardia globerula. Nocardia globerula NHB-2 cells grown on nutrient broth supplemented with 1% glucose and 0.1% yeast extract exhibited nitrile hydratase–amidase activity specific for saturated aliphatic nitriles or amide, while addition of acetonitrile in nutrient broth yielded cells with nitrile hydratase–amidase that in addition to saturated aliphatic nitriles–amide also hydrolyzed aromatic amide. Nocardia globerula NHB-2 cultivated on nutrient broth containing propionitrile exhibited nitrilase activity that hydrolyzed aromatic nitrile and unsaturated aliphatic nitrile. Under optimum fermentation conditions, each pellet developed as a round mass of mycelium of uniform consistency during the first 24 hours of the fermentation (mash sparged with air); subsequently (mash sparged with oxygen), a dense crust of growth formed at the periphery of the pellet and autolysis of cells at the center began. At the end of fermentation (140 hours), the pellet consisted of a shell of mycelium occupying less than 50% of the pellet volume. Changes in bacterial numbers and chemical composition of the oils were monitored using a plate count and chromatographic technique respectively. Populations induced under psychrophilic conditions readily metabolized similar quality oils under mesophilic conditions. Mesophilic populations, however, only showed a limited metabolic capability on similar quality oils under psychrophilic conditions. Gram-negative rods were predominant in all the populations obtained under these experimental conditions. The ability of the mixed populations to use crude oil as a sole carbon source was dependent not only on the composition and amount of the n-saturate fraction but also on that of the asphaltene and NSO (i.e. nitrogen-, sulfur-, and oxygen-containing) fraction. Growth on an oil which lacked a normal n-alkane component indicated that the aromatic fraction of oil was capable of sustaining bacterial growth. Oil quality and temperature of incubation affected the generic composition of populations obtained which would use crude oil. Our working hypothesis was that the rhizosphere effect would be strongest on fast-growing aerobic heterotrophs, while there would be little or no rhizosphere effect on oligotrophic and other slow-growing microorganisms. Culturable bacteria and fungi had larger population densities in the rhizosphere than in bulk soil. Communities were characterized by soil fatty acid analysis and by substrate utilization assays for bacteria and fungi. Fatty acid analysis revealed a very strong soil effect but little plant effect on the microbial community, indicating that the overall microbial community structure was not affected by the rhizosphere. There was a strong rhizosphere effect detected by the substrate utilization assay for fast-growing aerobic heterotrophic bacterial community structure, with soil controls and rhizosphere samples clearly distinguished from each other. There was a much weaker rhizosphere effect on fungal communities than on bacterial communities as measured by the substrate utilization assays.
